Earlier years

     Misao's father Okashira, was the original leader of the Oniwbanshuu. When he died, Misao was given over to be raised by the rest of the clan, and Aoshi became its leader. Misao idolizes Aoshi and has a big crush on him, constantly begging him to play with her and each her to fight. Misao was trained by Aoshi, Hannya (another member) and Okina. When Misao was about six or eight years old however, Aoshi and the other Oniwabanshuu left Kyoto to participate in the Bakumatsu no Douran war, and was gone for a long time. When Misao reached sixteen years of age, she embarked on a quest to look for her Oniwabanshuu team members Aoshi, Hannya, Beshimi, Hyotouko and Shikijou. Unbeknownst to her, Hannya, Beshimi, Hyoutoko and Shikijou had died, and Aoshi vows to avenge them, blaming Kenshin Himura for their deaths.

First meeting : Kenshin vs. Misao

     On his way to Kyoto to find Shishio Makoto and his Jun Pon Gatana, Kenshin pauses to rest in a nearby forest. Hearing a commotion, he goes over to investigate, only to find a group of thugs surrounding a prostitute, whose face was covered with a long cloak. After being promised a huge sum of money to...er, aheh *^__^*, the prostitute whips the cloak away, only to reveal Misao Makimachi, who was only tricking the men for their money that they had stolen earlier. Disappointed at finding an underdeveloped child instead of a sexy woman, the band stalks away. Pride injured, Misao goes on to beat them up. While relishing in the money she just acquired, Misao is startled as Kenshin approaches closer. Deciding she may as well rob him too, Misao attacks, but Kenshin successfully fends her off and accidentally damaging the bag where her belongings were tucked in. Mortified, Misao demands reparation, and Kenshin offers her his sakaba sword. Misao however, seeing the reverse edged blade, deems it worthless, and Kenshin does his 'oro' parody. He allows Misao to travel with him.....
    ....until he discovers that Misao is part of the Oniwabanshuu clan led by Aoshi Shinomori, his enemy.
    Kenshin tries to lose Misao, but Misao doggedly follows him, believing that Kenshin can lead her to where Aoshi, Hannya and the other Oniwabanshuu are. Finally, Kenshin leaps over a precipice and lands on the other side, knowing that Misao can't make the jump. Misao however, tells Kenshin that she'd willingly die looking for her tewammates and leaps over the cliff, and would have fallen to her death if not for Kenshin rescuing her. Once she recovers, Kenshin reluctantly agrees to have Misao accompany him to Kyoto, much to Misao's joy.

First meeting : Misao and Saitoh Hajime, "Don't call me weasel!"

       On their way to Kyoto, Kenshin and Misao encounters a town being controlled by the Jun Pon Gatana and rescues a young boy. Here they also meet Saitoh Hajime, who takes one look at her, points and goes ("since Kaoru's Raccoon Girl, and Megumi's Fox Lady, this must be....") "......Weasel Girl." Naturally, this does not suit well with Misao, but she can't hurt him no matter what she does since Saitoh's a policeman, and far stronger than she was.

Back to Kyoto

       Arriving at Kyoto, Misao invites Kenshin to stay at the Oniwabanshuu quarters. Here, Kenshin meets Okina and the rest of the Oniwabanshuu who were left behind with Misao (who eventually forms the new Oniwabanshuu). Kenshin leaves to search for Shishio again however, in a short while. In the meantime, Misao bumps into Kaoru and Yahiko around the Kyoto shops. Realizing that Kenshin was the man Yahiko and Kaoru were looking for, Misao invites them back home.

Betrayal

        Aoshi comes back to Kyoto, but he isn't the same man, now working with the Jun Pon Gatana to seek revenge against Kenshin. He comes back to kill Okina, who would be next in line as Oniwabanshuu leader, and nearly succeeds. Misao arrives just in time to see the final blow, and is stunned, not wanting to believe this is her Aoshi-sama. As he leaves, Aoshi tells Misao to forget her, which breaks her heart all the more. Later on, as they attend to the injured Okina, Misao pushes aside her feelings for Misao and the deaths of the orignal Oniwabanshuu, and announces that she will assume leadership of the Oniwabanshuu. Unfortunately, the others think she's joking at first.

Oniwabanshuu vs. Jun Pon Gatana

         Kenshin and Sanosuke leaves to battle Shishio Makoto (Saitoh Hajime joins them along the way), while many Jun Pon Gatana members leave Shishio's to destroy the Oniwabanshuu dojo. Among the fights were Misao versus Kamatari Honjo and Yahiko and Kaoru versus Flying Hennya. However, Misao freaks out when she learns Kamatari Honjo is a crossdresser (and because Kamatari showed her some of his anatomy that a woman doesn't have, to prove it^^;), and Yahiko insists that he can take Flying Hennya all by himself, and that Kaoru should help Misao. In the fight, Misao is heavily injured, but Hannya suddenly appears before her in some sort of hallucination, urging her not to give up. Encouraged, Misao fights back again, distracting Kamatari enough that Kaoru can get close and break his kneww, ending the fight. Later on, Kamatari wants to kill himself, but Misao stops him by knocking him unconscious.

Aftermath

         Kenshin and Sanosuke returns victorious (apparently, Saitoh died when the Jun Pon Gatana headquarters collapsed) and with them is Aoshi.
        (Aoshi had fought Kenshin again in the Jun Pon Gatana HQ, and Kenshin tells Aoshi that he promised Misao he would bring him back home, except that the man fighting him wasn't the Aoshi that Misao knew, therefore he'd be unable to keep his promise. They fight, and Aoshi finally realizes his mistake a little before Kenshin defeats him. He later on helps Kenshin, Saitoh and Sano fight  Shishio)
         Misao is overjoyed and wants to see Aoshi fast, but Okina says Aoshi is meditating at the moment and that she shouldn't disturb him. Later, Kenshin, Sanosuke, Kaoru and Yahiko leaves Kyoto and goes back to Tokyo.

Anime

          In the anime, Misao appears once more to accompany Yutaro (Kaoru's former student now studying to be a doctor) and the Kenshin - gumi to look for a miracle herb reputed to cure any sickness or disease, and to keep it from the clutches of the Black Knights, who wants to use it to control the world. With them is the woman leader of the Sorata ninjas, a group in which the Oniwabanshuu had been keeping a close eye on. They succeed, and Misao accompanies them back to Tokyo, where they bid Yutaro and his mentor/doctor farewell, as they leave for Germany.
          The other Oniwabanshuu arrives in Tokyo to accompany Misao back to Kyoto, and with them is Aoshi. Misao is overjoyed to see him, until she figures out that Aoshi as there not to see her, but because Okina commanded him to. Misao refuses to go back to Kyoto, and closets herself in one of the rooms of the Kamiya dojo. She is comforted by Ochika, Omasu (both Oniwabanshuu female members) Kaoru and Megumi, saying that Aoshi wouldn't obey anything if he didn't want to do it himself either. A little ashamed, Misao decides to go back to Kyoto, but Kaoru proposes they go tomorrow, and they look around Tokyo today instead.
           Misao, Megumi, Kaoru, Ochika and Omasu spends the day sightseeing in Tokyo, with Sano and Yahiko spying on them, having come to the conclusion earlier that they were being left out on the fun. Aoshi remains behind to share a tea ceremony with Kenshin. Watching Suzume and Ayume play with Kenshin, Aoshi couldn't help but remember how he used to do the same with Misao when she was younger. Kenshin points out, quite slyly, that Misao isn't a kid anymore. Aoshi reflects on that.
           Coming back from their trip (with an exhausted Sano and Yahiko not far behind them) Misao presents Aoshi with a gift; a paper ball he used to give her when she was younger, that she was a woman now. She shyly suggests that she and Aoshi  should talk about that further, and Aoshi agrees. Later on, they leave for Kyoto, and that's the last anyone would see of them.

Manga : Revenge Arc (this does not appear in the anime TV series)

           Misao and Aoshi arrives in Tokyo to visit the Kamiya Dojo, only to discover that Kaoru had been killed by Enishi Yukishiro, whose deceased sister Tomoe had been Kenshin's dead wife. Enishi blames Kenshin for Tomoe's death, and takes revenge by killing the woman Kenshin loves. Kenshin takes to the slums to live, and Yahiko and Misao runs about trying to discover what really happened to Kaoru.
